目的:研究阳和平喘颗粒对哮喘模型大鼠的保护作用及其可能的作用机制。方法:将60雄性只大鼠随机分为正常对照组、模型组、阳和平喘颗粒(25.92、12.96、6.48g/kg)组、桂龙咳喘宁胶囊(1.62g/kg)组。试验开始第一天,大鼠腹腔注射10%卵清蛋白(OVA)氢氧化铝生理盐水溶液致敏,第8天再次致敏,正常对照组注射等量生理盐水。于初次致敏第15天起,除正常组外,其余各组大鼠用含1%OVA的生理盐水超声雾化激发20min,隔天一次,共7次,复制大鼠哮喘模型。实验第15天起,给药组分别灌胃给予相应的药物,治疗2周,正常组和模型组给予等容积溶媒。实验结束后,分别采用硝酸还原酶法和放射免疫法测定血清中一氧化氮(NO)、内皮素-1(ET-1)的含量,HE染色法观察哮喘大鼠肺组织病理改变,免疫组化法观察诱导型一氧化氮合酶(iNOS)在肺组织中的表达。结果:与正常组比较,模型组大鼠出现明显的哮喘症状,肺组织及周围有大量炎性细胞浸润,血清中NO、ET-1含量显著升高,肺组织中iNOS的表达水平明显高于正常对照组;与模型组相比,阳和平喘颗粒(25.92、12.96g/kg)组哮喘大鼠哮喘症状及肺组织炎症反应明显减轻,大鼠血清中NO、ET-1的含量及肺组织iNOS活性显著降低。结论:阳和平喘颗粒对OVA介导的大鼠哮喘模型的保护作用可能与其对ET-1、NO、iNOS的调节有关。
OBJECTIVE: To study the protective effect of Yang Hechuan asthma on asthmatic rats and its possible mechanism. Methods: 60 male rats were randomly divided into normal control group, model group, Yang Pingchuan group (25.92, 12.96, 6.48g/kg) group and Guilong Kechuanning capsule (1.62g/kg) group. On the first day of the experiment, rats were given an intraperitoneal injection of 10% ovalbumin (OVA) in aqueous aluminum hydroxide solution and sensitized on the 8th day. The normal control group was injected with the same volume of physiological saline. On the 15th day after the initial sensitization, except for the normal group, the remaining rats in each group were ultrasonically atomized with physiological saline containing 1% OVA for 20 minutes, once every other day for 7 times, and the rat asthma model was duplicated. On the 15th day of the experiment, the drug administration group was given intragastric administration of the corresponding drug, and it was treated for 2 weeks. The normal group and the model group were given an equal volume of vehicle. After the end of the experiment, the levels of nitric oxide (NO) and endothelin-1 (ET-1) in serum were measured by nitrate reductase method and radioimmunoassay respectively. The pathological changes of lung tissue in asthmatic rats were observed by HE staining. The expression of inducible nitric oxide synthase (iNOS) in lung tissue was observed by chemical method. RESULTS: Compared with the normal group, the rats in the model group had obvious symptoms of asthma. There were a large number of inflammatory cells infiltrated in and around the lung tissues. The contents of NO and ET-1 in the serum were significantly increased, and the expression level of iNOS in the lung tissue was significantly higher than that in the normal group. Compared with the model group, compared with the model group, the symptoms of asthma and the inflammation of the lung tissue in the asthmatic rats of the Yang and Pingchuan granules (25.92, 12.96 g/kg) group were significantly reduced, and the contents of NO, ET-1 in the serum of the rats and lung tissue were significantly reduced. iNOS activity was significantly reduced. Conclusion: The protective effect of Yangheaichuan Granules on OVA-mediated rat asthma model may be related to the regulation of ET-1, NO and iNOS.
