目的:探讨腺苷能否诱导急性冠状动脉综合征(ACS)患者外周血单核细胞中血管内皮生长因子(VEGF)表达。方法:分别采取80例ACS患者发病后第1天的外周静脉血,随机分为4组,每组20例,对外周血单核细胞进行分离、培养,其中3组分别加入腺苷10μg、100μg、1000μg培养6 h、12 h和24 h。用酶联免疫吸附法测定VEGF浓度。结果:ACS患者VEGF水平在腺苷10μg组与对照组比差异无统计学意义(P>0.05)。VEGF水平在腺苷100μg组和1 000μg组均显著高于对照组(P<0.05或P<0.01),且与腺苷浓度和作用时间成正比。结论:腺苷能诱导ACS患者外周血单核细胞中血管VEGF表达且与浓度和作用时间成正比。
AIM: To investigate whether adenosine can induce the expression of vascular endothelial growth factor (VEGF) in peripheral blood mononuclear cells in patients with acute coronary syndrome (ACS). Methods: Peripheral venous blood was collected on the first day after onset of 80 patients with ACS. The peripheral venous blood was randomly divided into 4 groups (n = 20 in each group). Peripheral blood mononuclear cells were isolated and cultured. Addition of adenosine 10μg, 100μg , 1000μg cultured 6 h, 12 h and 24 h. The concentration of VEGF was measured by enzyme-linked immunosorbent assay. Results: There was no significant difference in VEGF level between adenosine 10μg group and control group (P> 0.05). The level of VEGF in adenosine 100μg group and 1 000μg group was significantly higher than that in control group (P <0.05 or P <0.01), and was proportional to adenosine concentration and time. Conclusion: Adenosine can induce the expression of vascular VEGF in peripheral blood mononuclear cells of patients with ACS, which is proportional to the concentration and duration of action.
