目的:应用Box-Behnken试验设计,优选超高压提取法提取连翘中连翘酯苷A的最佳工艺。方法:在单因素试验的基础上,采用3因素3水平Box-Behnken试验设计对连翘中连翘酯苷A的超高压提取工艺参数进行优选。以连翘酯苷A提取率为指标进行试验,考察液固比、提取压力、提取时间等因素对提取工艺的影响。结果:超高压提取连翘中连翘酯苷A的最佳工艺条件为液固比70 ml·g-1、压力151 MPa、提取时间114 s,此提取条件下连翘酯苷A的提取率为13.15 mg·g-1。结论:超高压提取作为一种新兴的中药提取技术,具有提取率高、提取温度低、耗时短、能耗少等优点,为连翘酯苷A提取提供一个全新的方法。
Objective: To apply Box-Behnken experimental design and optimize extraction of Forsythiaside A from Forsythia suspensa by ultra-high pressure extraction. Methods: Based on the single-factor experiment, the optimum conditions for the extraction of forsythiaside A from Hypericum perforatum were optimized by the Box-Behnken test design of 3-factor-3. Forsythiaside A extraction rate as an indicator of the test, liquid-solid ratio, extraction pressure, extraction time and other factors on the extraction process. Results: The optimum extraction conditions of Forsythiaside A in Forsythia suspensa Forsythiae were: liquid-solid ratio of 70 ml · g-1, pressure of 151 MPa and extraction time of 114 s. The extraction rate of Forsythiaside A 13.15 mg · g-1. Conclusion: As a new extraction technology of Chinese herbal medicine, extra-high pressure extraction has the advantages of high extraction rate, low extraction temperature, short time-consuming and low energy consumption, which provides a new method for the extraction of Forsythoside A.
