目的 通过检测肾癌相关新基因GYLZ RCC1 8(基因号 :BE82 51 3 3 )在肾癌中的表达特点及其对肾癌细胞生长、增殖的影响 ,探讨GYLZ RCC1 8基因的功能及其在肾癌发生发展中的作用。方法 应用逆转录 聚合酶链式反应 (RT PCR)的方法检测肾癌相关基因GYLZ RCC1 8在3 1例肾癌标本中的表达特点 ;观察GYLZ RCC1 8反义寡核苷酸对肾癌细胞生长、增殖活性、形态学的影响。结果 GYLZ RCC1 8基因在肾癌组织特异表达 ,正常肾组织较肾癌组织低表达或不表达 ,两者表达量差异为 1~ 9倍。GYLZ RCC1 8基因的表达在高分级、高分期肾癌明显高于低分级、低分期肾癌 ;导入GYLZ RCC1 8反义寡核苷酸后 ,可明显抑制癌细胞的生长和增殖活性 ,并减少GRC 1细胞的核分裂。结论 GYLZ RCC1 8是肾癌组织相对较特异表达的新基因 ,它的高表达与肾癌的发生发展以及生长、增殖密切相关 ,它的成功克隆为阐明肾癌发生发展的分子生物学机制开辟了新的途径 ,为今后肾癌的特异诊断 ,基因治疗提供了新的理论依据。
Objective To detect the expression of GYLZ RCC1 8 (Gene No .: BE82 51 3 3) in renal cell carcinoma and its effect on the growth and proliferation of renal cell carcinoma (RCC), and to explore the function of GYLZ RCC1 8 gene and its role in kidney The role of cancer in the development. Results The gene of GYLZ RCC1 8 was specifically expressed in renal cell carcinoma. The expression of GYLZ RCC1 8 gene was lower in normal renal tissues than in renal carcinoma tissues, with a difference of 1 to 9 times. GYLZ RCC1 8 gene expression in high grade, high staging renal cell carcinoma was significantly higher than low grade, low staging renal cell carcinoma; GYLZ RCC1 8 antisense oligonucleotide can significantly inhibit the growth and proliferation of cancer cells and reduce Mitosis of GRC-1 cells. Conclusions GYLZ RCC1 8 is a novel gene with relatively specific expression in renal cell carcinoma. The high expression of GYLZ RCC1 8 is closely related to the development, growth and proliferation of renal cell carcinoma. The successful cloning of GYLZ RCC1 8 opens up the molecular biological mechanism for the development of renal cell carcinoma New way for the future diagnosis of kidney cancer, gene therapy provides a new theoretical basis.
