目的:建立测定齐多夫定含量的新方法。方法:以甲基丙烯酸为功能单体,三甲氧基丙基三甲基丙烯酸酯为交联剂,合成抗HIV药物齐多夫定分子印迹聚合物,将其用于固相萃取,通过渗漏实验选择功能单体并考察了最大吸附量和最佳上样条件。结果:当以浓度为4.5 mmol·L~(-1),pH=7的缓冲溶液上样时,最大吸附量达到59.7 mg·g~(-1)。并通过对上样、洗脱条件进行优化,成功地将齐多夫定从其结构类似物拉米夫定的混合物中分离出来。不同齐多夫定和拉米夫定加入量的回收率分别为101.6%~102.6%,103.4%~104.0%,RSD分别为1.6%~4.2%,1.4%~3.3%。结论:该方法快速、直观,可用于药物的分离与富集。
Objective: To establish a new method for determination of zidovudine. Methods: Zidovudine molecularly imprinted polymer was synthesized by using methacrylic acid as functional monomer and trimethoxypropyltrimethacrylate as cross-linking agent, which was used for solid-phase extraction Experimental selection of functional monomer and investigated the maximum adsorption capacity and the best loading conditions. Results: The maximum adsorption capacity was 59.7 mg · g ~ (-1) when loaded with buffer solution of 4.5 mmol·L ~ (-1) and pH = 7. The zidovudine was successfully separated from the mixture of its structural analogs lamivudine by optimizing loading and elution conditions. The recoveries of different doses of zidovudine and lamivudine were 101.6% -102.6% and 103.4% -104.0%, respectively. The RSDs were 1.6% -4.2% and 1.4% -3.3%, respectively. Conclusion: The method is rapid and intuitive and can be used for the separation and enrichment of drugs.
