目的:比较通过卵巢局部注射和尾静脉注射两种不同的移植方式,脐带间充质干细胞(UCMSCs)在卵巢损伤小鼠体内的分布、迁移和增殖,探讨最佳移植途径。方法:取8周龄ICR雌性小鼠12只,采用环磷酰胺(CTX)200 mg/kg+白消安(BUS)30 mg/kg一次性腹腔注射方式建立卵巢损伤小鼠模型;采用慢病毒转染途径构建标记有荧光素酶(Luc)的UCMSCs;将卵巢损伤小鼠随机分为两组,分别采用尾静脉移植(2×10n 6个细胞)和单侧卵巢局部移植(2×10n 5个细胞),移植后采用小动物活体成像(IVIS)技术示踪UCMSCs在小鼠体内的分布、迁移及增殖。组间比较采用n t检验。n 结果:化疗药物处理后1周小鼠血清FSH水平[(17.971±0.311) μg/L]显著高于化疗药物处理前[(14.420±0.622) μg/L]、化疗药物处理后1周小鼠血清E2水平[(320.321±8.682) ng/L]显著低于化疗药物处理前[(175.383±19.400) ng/L],差异有统计学意义(n t=5.106、4.800,n P<0.05);卵巢局部移植Luc-UCMSCs后在小鼠卵巢部位可观察到荧光信号(d1:2.61×10n 5±0.33、d2:1.35×10n 5±0.23、d3:3.09×10n 5±0.29、d4:4.12×10n 5±0.43),其余部位未见荧光信号,并且在移植第2天开始荧光信号逐渐增强,差异有统计学意义(n t=7.011、8.311、4.562,n P<0.05),第5天信号消失,而尾静脉移植组小鼠在移植后7 d内各部位均未见到明显荧光信号。n 结论:UCMSCs通过卵巢局部注射可以在小鼠受损的卵巢局部聚集并存活,与尾静脉注射比较,卵巢局部注射可能会取得更好的治疗效果。“,”Objective:To find the optimal transplantation route by comparing the local ovarian injection and tail vein injection, and the distribution, migration and proliferation of umbilical cord mesenchymal stem cells (UCMSCs) in mice models of ovarian injury via then in vivo imaging system.n Methods:A total of 200 mg/kg cyclophosphamide and 30 mg/kg busulfan were injected intraperitoneally to establish the mice models of ovarian injury. Lentiviral transfection was done to construct UCMSCs labeled with luciferase (Luc). The mice models of ovarian injury were randomly divided into two groups: tail vein injection group (2×10n 6 cells) and unilateral ovarian local injection group (2×10n 5 cells). The distribution, migration and proliferation of UCMSCs in the mice models of ovarian injury were observed by then in vivo imaging system.n Results:Luc-UCMSCs injected unilaterally into the ovary showed a gradual increase in the fluorescence signal on the second day to the fourth day after transplantation, and no fluorescence signal was found in the other parts of the body. On the fifth day after transplantation, the fluorescence signal disappeared. In the tail vein injection group, fluorescence signals were not observed in all parts of the body at 7th day after transplantation.Conclusion:UCMSCs can aggregate and proliferate in damaged ovarian organs of mice through local ovarian injection. The local ovarian injection may achieve better therapeutic effect than the tail vein injection.
