研制了一种高精度光纤生物传感器,以用于低浓度葡萄糖的检测。传感器探头组装有荧光氧敏感膜和葡萄糖氧化酶(GOD)膜,获得了GOD在醋酸纤维素(CA)膜上的最佳固定条件:高碘酸钠浓度为0.2 mol/L,反应时间为1 h;乙二胺的浓度为0.1 mol/L,反应时间为2.5 h;戊二醛的浓度为2%,反应时间为90 min;GOD的浓度为25 mg/mL,固定时间为8 h。采用锁相放大技术,通过测定传感器探头滞后相移变化来测定葡萄糖的浓度。传感器的检测范围为1~10mg/dL,响应时间小于1 min,最佳使用pH为6.0,最佳使用温度为32℃,并具有较好的重复性和稳定性。
A high-precision fiber optic biosensor was developed for the detection of low concentrations of glucose. The sensor probe was assembled with a fluorescence oxygen sensitive membrane and a glucose oxidase (GOD) membrane to obtain the optimal immobilization conditions of GOD on a cellulose acetate (CA) membrane: the sodium periodate concentration was 0.2 mol / L and the reaction time was 1 h; the concentration of ethylenediamine was 0.1 mol / L, the reaction time was 2.5 h; the concentration of glutaraldehyde was 2%, the reaction time was 90 min; the concentration of GOD was 25 mg / mL and the immobilization time was 8 h. The use of phase-locked amplification technology, by measuring the sensor probe delay phase shift to determine the concentration of glucose. Sensor detection range of 1 ~ 10mg / dL, response time of less than 1 min, the best use of pH 6.0, the best temperature of 32 ℃, and has good repeatability and stability.
