INTRODUCTION Electron microscopes with an acceleration voltage of 70,000 volts and a magnification of 12 times have been produced by E. Rusk and M. Knoll of the High-Voltage Laboratory of German University of Technology since 1932, for half a century. Now the electron microscope has a high resolution, high magnification, multi-purpose performance. It has been widely used in many fields of science and technology. Plant fiber cell wall structure of various levels, lignin, hemicellulose, polysaccharide distribution, the use of optical microscopy is powerless. The scientists concerned long ago hoped to directly observe the internal microscopic world of plant fibers,
