目的 研究当归对脑缺血半暗带细胞凋亡的影响。方法 采用大鼠局灶性脑缺血再灌流模型 ,大脑中动脉阻塞 2小时 ,再灌流 48小时 ,用TUNEL法和免疫组织化学染色法分别检测使用当归后缺血半暗带凋亡细胞和凋亡相关基因Bcl- 2、Bax蛋白表达。结果 与对照组比较 ,当归治疗组缺血半暗带的凋亡细胞显著减少 (P <0 .0 1) ;Bcl- 2的表达显著增加 (P <0 .0 1) ,Bax的表达无显著变化 (P >0 .0 5 )。结论 当归可能通过促进Bcl- 2在脑缺血后的表达对半暗带的细胞凋亡产生抑制作用。
Objective To study the effect of Angelica sinensis on apoptosis of cerebral ischemic penumbra. Methods The rat model of focal cerebral ischemia and reperfusion was used. The middle cerebral artery was occluded for 2 hours and then perfused for 48 hours. The apoptotic cells in the ischemic penumbra were detected by TUNEL and immunohistochemical staining. The expression of Bcl-2 and Bax proteins related to death. Results Compared with the control group, apoptotic cells in the ischemic penumbra were significantly decreased in the Angelica treatment group (P < 0.01); Bcl-2 expression was significantly increased (P <0.01); Bax expression was not significant. Change (P > 0. 0 5). Conclusion Angelica may inhibit the apoptosis of the penumbra by promoting the expression of Bcl-2 after cerebral ischemia.
