[目的]探讨TGF-β1对胃癌细胞株SNU-601/WT和耐药胃癌细胞株SNU-601/cis2细胞生长的影响及与c-myc,P 53蛋白之间的相关关系.[方法]利用MTT法观察TGF-β1对胃癌细胞生长的抑制情况;用免疫细胞化学方法观察c-myc,P 53蛋白的表达情况.[结果]实验组SNU-601/WT和SNU-601/cis 2N胞的生存率逐渐下降,具有时间依赖性,与对照组相比均有非常显著性差异;48,72 h时耐药胃癌细胞株的生存率比胃癌细胞株明显降低;实验组c-myc,P 53蛋白表达均明显降低.[结论] TGF-β1可抑制SNU-601/WT和SNU-601/cis 2细胞的生长,其机制与抑制c-myc,P 53蛋白的表达有关.
[Objective] To investigate the effect of TGF-β1 on the growth of gastric cancer cell line SNU-601/WT and drug-resistant gastric cancer cell line SNU-601/cis2 and the correlation with c-myc, P 53 protein. The inhibitory effect of TGF-β1 on the growth of gastric cancer cells was observed by MTT method; the expression of c-myc and P 53 protein was observed by immunocytochemistry. [Results] The results of SNU-601/WT and SNU-601/cis 2N cells in experimental group The survival rate decreased gradually and was time-dependent. Compared with the control group, there was a very significant difference. The survival rate of resistant gastric cancer cell lines at 48 and 72 hours was significantly lower than that of gastric cancer cell lines. The experimental group c-myc, P53 Protein expression was significantly reduced. [Conclusion] TGF-β1 can inhibit the growth of SNU-601/WT and SNU-601/cis 2 cells, and its mechanism is related to the inhibition of c-myc, P 53 protein expression.
