过氧化氢清除剂对H_2O_2处理下中国石竹幼苗不同器官抗氧化系统的影响

以中国石竹为试材,采用25mmol·L~(-1)二甲基硫脲(dimethylthiourea,DMTU)和3mmol·L~(-1)二苯基氯化碘盐(diphenyleneiodonium chloride,DPI)对其幼苗预处理2h后再置于5mmol·L~(-1) H_2O_2中处理6h,测定DMTU和DPI预处理对H_2O_2处理下中国石竹幼苗不同器官的H_2O_2和O·2含量以及相关清除酶活性的方法,探究减少H_2O_2含量对氧化胁迫下石竹幼苗生理活性的影响,以期为石竹抗逆栽培提供理论依据。结果表明:25mmol·L~(-1) DMTU和3mmol·L~(-1) DPI预处理降低了正常条件下叶片和根的H_2O_2和O·2含量,且在根中达显著差异水平(P<0.5);5mmol·L~(-1)的外源H_2O_2处理显著增加了叶中H_2O_2含量和根中O·2含量(P<0.5),25mmol·L~(-1) DMTU和3mmol·L~(-1) DPI预处理则降低了5mmol·L~(-1) H_2O_2处理下叶片和根部的H_2O_2和O·2含量;5mmol·L~(-1)的外源H_2O_2处理显著增加了叶片SOD活性和根部SOD、APX活性(P<0.5),25mmol·L~(-1) DMTU和3mmol·L~(-1) DPI预处理降低了5mmol·L~(-1)H_2O_2处理下石竹叶中SOD、APX活性,且25mmol·L~(-1) DMTU预处理显著降低了根部的APX活性(P<0.5),表明降低H_2O_2和O·2含量可降低相应清除酶的活性。因此,外施DMTU和DPI能不同程度缓解H_2O_2处理导致的内源H_2O_2和O·2含量的积累,且25mmol·L~(-1) DMTU清除H_2O_2的效果比3mmol·L~(-1) DPI好。 Dianthus chinensis was used as test material, and 25 mmol·L -1 dimethylthiourea (DMTU) and 3 mmol·L -1 diphenyleneiodonium chloride (DPI) Seedlings were pretreated for 2h and then treated with 5mmol·L -1 H 2 O 2 for 6h. The contents of H_2O_2 and O_2 in different organs of Dianthus chinensis seedlings under H_2O_2 treatment and the activities of related enzymes were measured by DMTU and DPI pretreatment , To explore the effect of decreasing H 2 O 2 content on the physiological activity of Dianthus chinensis seedlings under oxidative stress so as to provide a theoretical basis for the resistance cultivation of Dianthus. The results showed that pretreatment with 25 mmol·L -1 DMTU and 3 mmol·L -1 DPI decreased the contents of H 2 O 2 and O 2 in leaves and roots under normal conditions and reached significant differences in root (P <0.5). Exogenous H 2 O 2 with 5 mmol·L -1 significantly increased the contents of H 2 O 2 and O 2 in root (P <0.5), 25 mmol·L -1 DMTU and 3 mmol·L ~ (-1) DPI pretreatment decreased the content of H_2O_2 and O_2 in leaves and roots under the treatment of 5 mmol·L -1 H_2O_2. Exogenous H_2O_2 with 5 mmol·L -1 significantly increased SOD activity and root activity of SOD, APX activity (P <0.5), 25mmol·L -1 DMTU and 3mmol·L -1 DPI decreased the content of Pholiota leaves (P <0.05). The results showed that decreasing the content of H 2 O 2 and O 2 could reduce the activity of the corresponding scavenging enzymes. Therefore, the application of DMTU and DPI could alleviate the accumulation of endogenous H 2 O 2 and O 2 caused by H 2 O 2 treatment to a certain extent, and the effect of 25 mmol·L -1 DMTU in removing H 2 O 2 was better than that of 3 mmol·L -1 DPI it is good.
