目的:建立高糖输注诱导胰岛β细胞增殖的大鼠模型,为研究胰岛β细胞增殖的机制提供研究平台。方法:SD大鼠颈静脉插管后用输液泵连续输注72h的50%葡萄糖和0.45%的NaCl,用BrdU掺入和标记增殖蛋白ki67检测大鼠胰岛β细胞增殖情况,并检测增殖相关基因的表达。结果:输注50%葡萄糖组的大鼠与NaCl组比较胰岛β细胞BrdU和ki67阳性细胞明显增多,增殖相关基因ccnd2,c-myc表达增高,P27降低。结论:通过持续的高糖输注72h可建立胰岛β细胞增殖的大鼠模型。
OBJECTIVE: To establish a rat model of pancreatic β cell proliferation induced by high glucose infusion and provide a research platform for studying the mechanism of pancreatic β cell proliferation. Methods: Sprague - Dawley rats were injected with 50% glucose and 0.45% NaCl after infusion for 72 hours in the jugular vein. Proliferation of pancreatic islet β cells was detected by BrdU incorporation and labeling of proliferation protein ki67. Proliferation - expression. Results: BrdU and ki67 positive cells in pancreatic β cells were significantly increased in rats fed 50% glucose compared with those in NaCl group. The expression of proliferation related genes ccnd2 and c-myc were increased and P27 was decreased. Conclusion: A rat model of islet β cell proliferation can be established by continuous high glucose infusion for 72 hours.
