目的:建立牛樟芝水溶性成分的HPCE-DAD指纹图谱分析方法。方法:采用高效毛细管电泳法进行色谱分离,以60mmol/L硼砂(p H值9.3)为运行缓冲液,分离电压为22k V,检测波长为260nm,压力进样为50mbar×6s,毛细管温度为28℃。以腺苷为参照物,测定其指纹图谱,并作模糊聚类分析和相似度评价。结果:初步建立了以10个共有峰为特征指纹信息的牛樟芝水溶性成分的HPCE-DAD指纹图谱,相似度较高,色谱分离度较好,达到了中药指纹图谱的技术要求。结论:方法准确可靠,重现性好,可用于牛樟芝内在质量的评价和控制。
Objective: To establish a HPLC-DAD fingerprint method for the analysis of water-soluble components of Antrodia camphorata. Methods: Chromatographic separation was performed by high performance capillary electrophoresis with 60 mmol / L borax (pH 9.3) as the running buffer. The separation voltage was 22 kV, the detection wavelength was 260 nm, the pressure was 50 mbar × 6 s and the capillary temperature was 28 ℃. Using adenosine as a reference, the fingerprinting was determined, and fuzzy clustering analysis and similarity evaluation were performed. Results: The HPCE-DAD fingerprints of water-soluble constituents of Antrodia camphorata with 10 common peaks were preliminarily established, with high similarity and good chromatographic resolution, which met the technical requirements of fingerprinting. Conclusion: The method is accurate, reliable and reproducible. It can be used to evaluate and control the intrinsic quality of Antrodia camphorata.
