目的:探究解毒胶囊对乳腺增生大鼠乳腺组织中VEGF和b FGF表达的影响。方法:选取50只12周龄SD雌性未孕大鼠随机分为5组(正常组、模型组、方药组、三苯氧胺组、乳癖消组),每组各10只。对于模型组和3个治疗组进行乳腺增生模型的复制,造模成功后,方药组、三苯氧胺组、乳癖组分别给予解毒胶囊、三苯氧胺、乳癖消的水溶液灌胃。治疗4周后,光镜下观察各组大鼠乳腺组织的病理变化。用免疫组化和QPCR检测各组大鼠乳腺组织VEGF和b FGF表达情况。结果:光镜下可见乳腺增生大鼠模型复制成功,且解毒胶囊有治疗作用。免疫组化结果显示正常组乳腺组织内VEGF、b FGF呈阴性或弱阳性表达;模型组的VEGF、b FGF呈强阳性表达,显著高于正常组(P<0.01);而治疗三组的VEGF、b FGF阳性表达明显减轻,显著低于模型组(P<0.01);方药组乳腺组织内VEGF、b FGF的表达要明显低于乳癖消组(P<0.01、P<0.05),与三苯氧胺组相比无统计学差异。QPCR结果显示模型组与正常组相比VEGF和b FGF呈现高表达(P<0.01);治疗组(方药组、三苯氧胺组、乳癖消组)与模型组相比,VEGF和b FGF表达均减弱(P<0.01);其中方药组及三苯氧胺组比乳癖消组减弱明显(P<0.01),其比较结果均有统计学差异。而方药组与三苯氧胺组相比,结果没有统计学差异(P>0.05)。结论:解毒胶囊能够干预乳腺增生大鼠的乳腺组织VEGF和b FGF的表达情况,对乳腺增生有调节和治疗作用。
Objective: To investigate the effect of Jiedu capsule on the expression of VEGF and b FGF in mammary glands of rats with hyperplasia of mammary glands. Methods: Fifty female SD rats of 12 weeks old were randomly divided into 5 groups (normal group, model group, prescription drug group, tamoxifen group, Ruupoxiao group), 10 in each group. For the model group and the three treatment groups were replicated breast hyperplasia model, modeling success, the prescription drug group, tamoxifen group, milk addiction group were given detoxification capsules, tamoxifen, Rupixiao aqueous solution gavage. After 4 weeks of treatment, the pathological changes of breast tissue in each group were observed under light microscope. Immunohistochemistry and QPCR were used to detect the expression of VEGF and b FGF in mammary gland of rats in each group. Results: The model of hyperplasia of mammary glands can be successfully replicated in light microscope and the detoxification capsule has a therapeutic effect. The results of immunohistochemistry showed that VEGF and b FGF were negative or weakly positive in normal breast tissue. The expression of VEGF and b FGF in model group was significantly higher than that in normal group (P <0.01) (P <0.01). The expression of VEGF and bFGF in breast cancer tissue of the prescription group was significantly lower than that of Ruupoxiao group (P <0.01, P <0.05) There was no significant difference between the two groups. The results of QPCR showed that the expression of VEGF and b FGF were significantly higher in the model group than in the normal group (P <0.01). The expression of VEGF and b FGF in the treatment group (prescription group, tamoxifen group, Ruupoxiao group) (P <0.01). The prescription drug group and the tamoxifen group were significantly weaker than those of the milk addiction elimination group (P <0.01), the results of which were statistically different. However, there was no significant difference between the prescription group and the tamoxifen group (P> 0.05). Conclusion: Jiedu capsule can interfere with the expression of VEGF and b FGF in mammary gland tissue of rats with hyperplasia of mammary glands, and it can regulate and treat breast hyperplasia.
