【目的】观察细胞外热休克蛋白70(e HSP70)模拟创伤刺激内皮细胞后血管生成素2(Ang-2)及血管内皮钙黏蛋白(VEcadherin)的变化,探讨血必净注射液对血管内皮的保护作用。【方法】培养人脐静脉内皮细胞,检测e HSP70刺激0、2、4、8、16、32 h后细胞Ang-2、VE-cadherin的m RNA和蛋白表达量,找出最佳刺激时间,然后设空白对照组、e HSP70组(终浓度为0.1μg/m L)、血必净组(终浓度为50 mg/m L)、血必净+e HSP70组共4个实验组,检测最佳时间点各组上述指标改变。【结果】e HSP70刺激后Ang-2的m RNA及蛋白表达量增加并于4 h达到高峰,而VE-cadherin的m RNA和蛋白表达量经e HSP70刺激后降低,并于4 h达到最低值。各实验组干预4 h后,e HSP70组与血必净+e HSP70组细胞Ang-2的m RNA及蛋白表达量较空白对照组显著增高(P<0.05),而血必净+e HSP70组的表达量较e HSP70组显著下降(P<0.05);与空白对照组相比较,e HSP70组VE-cadherin的m RNA及蛋白的表达量显著降低(P<0.05),而血必净+e HSP70组及血必净组上述指标表达量均显著上升(P<0.05),其中尤以血必净组的表达增高最为显著(P<0.05)。【结论】血必净注射液通过调控e HSP70对Ang-2与VE-cadherin的表达影响,对创伤刺激的血管内皮细胞具有一定的保护作用。

The mRNA and protein expressions of Ang-2 in e HSP70 group and Xuebijing + e HSP70 group were significantly higher than those in blank control group (P <0.05) after 4 h of intervention, while those in Xuebijing + e HSP70 group (P <0.05). Compared with the blank control group, the expression of m RNA and protein of VE-cadherin in e HSP70 group was significantly lower than that in e HSP70 group (P <0.05) The HSP70 and Xuebijing groups above-mentioned indicators were significantly increased (P <0.05), especially Xuebijing group expression was the most significant (P <0.05). 【Conclusion】 Xuebijing injection can protect the vascular endothelial cells stimulated by trauma by regulating the expression of Ang-2 and VE-cadherin by e HSP70.
