目的将随机引物应用于黄热病毒核酸检测,为口岸快速准确地检测黄热病毒提供技术支持。方法采用随机引物和特异性引物进行逆转录,分别用自行设计的引物、探针和参考文献的引物、探针,用实时荧光PCR方法对6份黄热病毒阳性尿液样本进行黄热病毒核酸检测,并对检测结果进行比较。结果与特异性引物逆转录相比,随机引物逆转录的实时荧光PCR Ct值差别不显著(F值=0.11,P>0.05)。结论随机引物可直接应用于黄热病毒检测,也可在口岸传染病监测中发挥其他作用。
Objective To apply random primers to the detection of yellow fever virus nucleic acid and provide technical support for rapid and accurate detection of yellow fever virus at the port. Methods Random primers and specific primers were used for reverse transcriptase. Primers, probes and reference primers were designed and used respectively. Real-time PCR was performed on 6 samples of yellow fever virus positive urine samples for yellow fever virus nucleic acid Test, and compare the test results. Results Compared with the reverse transcription of specific primers, there was no significant difference (C> = 0.11, P> 0.05) between real-time PCR and RT-PCR. Conclusion Random primers can be directly applied to detect yellow fever virus, but also can play an other role in the monitoring of port infectious diseases.
