目的探讨分离部位是否影响牙周膜干细胞成骨能力。方法分离、培养牙根表面和牙槽窝内的牙周膜干细胞,并进行体外矿化能力检测、实时定量RT-PCR检测、ALP活性检测和体内动物实验。结果牙槽窝内的牙周膜干细胞人牙周膜干细胞矿化能力高于牙根表面的牙周膜干细胞。牙根表面的人牙周膜干细胞移植物显示出在CBB表面有牙骨质和牙周纤维样组织形成;而牙槽窝内的牙周膜干细胞移植物形成的大部分是类骨样结构。结论牙根表面牙周膜干细胞更适合重建牙骨质/牙周膜样结构,牙槽窝牙周膜干细胞更适合重建骨样结构。
Objective To investigate whether the isolated site affects the osteogenic potential of periodontal ligament stem cells. Methods The periodontal ligament stem cells in root surface and alveolar nest were isolated and cultured. The ability of mineralization in vitro, real-time quantitative RT-PCR, ALP activity and in vivo animal experiments were also studied. Results The periodontal ligament stem cells in periodontal ligament were longer than the periodontal ligament stem cells. Human periodontal ligament stem cell grafts on the root surface showed cementum and periodontal fibroid tissue formation on the surface of CBB; whereas the majority of periodontal ligament stem cell grafts formed in the alveolar fossa were osteoid-like structures. Conclusion Periodontal ligament stem cells are more suitable for reconstructing cementum / periodontal ligament-like structures. Periodontal ligament stem cells are more suitable for reconstructing bone-like structures.
