目的检测恶性黑素瘤组织中环氧化酶2(COX-2)与缺氧诱导因子-1α(HIF-1α)的表达,以明确其在恶性黑素瘤发生、发展中的临床意义。方法收集40例恶性黑素瘤和20例色素痣组织,采用免疫组化方法检测组织中COX-2与HIF-1α的表达并进行相关分析。结果 40例黑素瘤患者中33例(82.5%)COX-2(+),高于对照组(15%)(P<0.01)。黑素瘤患者中35例(87.5%)HIF-1α(+),高于对照组(10%)(P<0.01)。恶性黑素瘤患者组织中COX-2与HIF-1α表达存在正相关(r=0.324,P<0.05)。恶性黑素瘤HIF-1α表达在年龄、性别和部位分组中,差异不显著(P>0.05)。而恶性黑素瘤患者中病理分期高,且有淋巴结转移者则HIF-1α阳性率高(P<0.05)。结论 HIF-1α参与恶性黑素瘤的进展,可能与COX-2协同作用。
Objective To detect the expression of cyclooxygenase 2 (COX-2) and hypoxia-inducible factor-1α (HIF-1α) in malignant melanoma and to clarify its clinical significance in the occurrence and development of malignant melanoma. Methods 40 cases of malignant melanoma and 20 cases of nevus were collected. The expressions of COX-2 and HIF-1α were detected by immunohistochemistry and correlated with each other. Results 33 (82.5%) COX-2 (+) in 40 melanoma patients were higher than those in control group (15%) (P <0.01). 35 (87.5%) HIF-1α (+) in melanoma patients were higher than those in control group (10%) (P <0.01). There was a positive correlation between the expression of COX-2 and HIF-1α in malignant melanoma (r = 0.324, P <0.05). The expression of HIF-1α in malignant melanoma was not significantly different in age, sex and location group (P> 0.05). The malignant melanoma patients with high pathological stage, and those with lymph node metastasis HIF-1α positive rate (P lt; 0.05). Conclusions HIF-1α is involved in the progression of malignant melanoma and may play a synergistic role with COX-2.
