目的探讨免疫脂质化的多烯紫杉醇对LoVo细胞的放射增敏作用。方法制备偶联CEA抗体的多烯紫杉醇脂质体,测定抗体活性及细胞结合率,分析其对LoVo细胞体外放射增敏作用。结果多烯紫杉醇免疫脂质体平均粒径为:(213.0±17.4)nm、包封率为97.6%、较多烯紫杉醇脂质体处理的LoVo细胞荧光强度差异有统计学意义(P<0.01);偶联抗CEA单抗的免疫活性无明显改变;2nmol/L和100nmol/L多烯紫杉醇免疫脂质体均可提高LoVo细胞放射敏感性。SER_(D0)分别为1.70、1.78,而2nmol/L多烯紫杉醇脂质体对其放射敏感性的的影响不明显。SER_(D0)为1.02。结论多烯紫杉醇免疫脂质体可特异的与LoVo细胞结合并具有放射增敏作用。
Objective To investigate the radiosensitization effect of immunolipidated docetaxel on LoVo cells. Methods The docetaxel liposomes conjugated with CEA antibody were prepared and their activity and cell binding rate were measured. The radiosensitization of LoVo cells in vitro was analyzed. Results The average particle size of docetaxel immunized liposomes was (213.0 ± 17.4) nm, the entrapment efficiency was 97.6%. There was a statistically significant difference in the fluorescence intensity of LoVo cells treated with liposomes containing paclitaxel (P <0.01). The immunological activities of conjugated anti-CEA mAb did not change significantly. Immuno-liposomes with both 2nmol / L and 100nmol / L docetaxel could increase the radiosensitivity of LoVo cells. SER_ (D0) were 1.70 and 1.78, respectively. However, the effect of 2nmol / L docetaxel liposomes on radiosensitivity was not obvious. SER_ (D0) is 1.02. Conclusion Docetaxel immunoliposomes can specifically bind to LoVo cells and have radiosensitization.
