本文叙述一种将辣根过氧化物酶(HRP)与单链DNA交联以制备非同位素核酸杂交探针的新方法。HRP-单链DNA复合物可用于斑点杂交,经杂文并与HRP底物溶液作用后,可直接检出互补的DNA靶序列,而不需繁冗的细胞化学夹心法。本法可检出 1~4 pg的靶DNA。
This article describes a new method for the preparation of non-isotopic nucleic acid hybridization probes by cross-linking horseradish peroxidase (HRP) with single-stranded DNA. The HRP-single-stranded DNA complex can be used for dot blotting. After hybridization with HRP substrate solution, complementary DNA target sequences can be directly detected without the cumbersome cytochemical sandwich method. This method can detect 1 ~ 4 pg of target DNA.
