通过正交设计试验,探讨了超临界CO2法萃取北五味子果实中五味子醇甲的工艺条件,并建立了五味子醇甲的质量分数高效液相色谱测定方法。研究表明:最佳提取条件为萃取压力25MPa,萃取温度45℃,萃取时间60min,提取率可达到0.970%;以乙腈-水溶液为流动相,在Zorbax Eclipse XDB-C18柱进行高效液相色谱分析,平均回收率为98.64%,相对标准偏差为2.908%,检出限为0.02μg/L
Through the orthogonal test, the technological conditions of extracting Schisandra chinensis fruit from the fruit of Schisandra chinensis by supercritical CO2 were discussed. And the determination method of schisandrin A was established by mass fraction high performance liquid chromatography. The results showed that the optimal extraction conditions were as follows: extraction pressure 25MPa, extraction temperature 45 ℃, extraction time 60min, the extraction rate could reach 0.970%. High performance liquid chromatography (HPLC) on a Zorbax Eclipse XDB-C18 column with acetonitrile-water as mobile phase, The average recovery was 98.64%, the relative standard deviation was 2.908%, the detection limit was 0.02μg / L
