目的通过研究CD8~+CD28~+在胃癌肿瘤浸润淋巴细胞(TIL)上的表达及其与肿瘤生物学行为的关系和意义,探讨TIL功能受抑制的原因。方法采用流式细胞术检测胃癌TIL及胃癌患者和正常健康志愿者外周血淋巴细胞CD3、CD4、CD8、CD28的表达。结果胃癌TIL表面CD3~+、CD3~+CD4~+、CD3~+CD8~+、CD8~+CD28~+的表达均低于正常健康志愿者外周血和胃癌患者外周血中的表达,差异有统计学意义(P<0.05);胃癌TIL表面CD8~+CD28~+的表达与肿瘤浸润深度和肿瘤大小有关(P<0.05)。结论胃癌TIL的免疫抑制状态可能与CD8~+CD28~+低表达有关;CD8~+CD28~+在TIL上的表达可作为胃癌患者预后的一个参考指标。
OBJECTIVE: To investigate the expression of CD8 + CD28 + on gastric tumor infiltrating lymphocytes (TIL) and its relationship with tumor biological behavior, and to explore the possible reasons for the inhibition of TIL function. Methods Flow cytometry was used to detect the expression of CD3, CD4, CD8 and CD28 in peripheral blood lymphocytes of patients with gastric cancer TIL, gastric cancer and normal healthy volunteers. Results The expression of CD3 ~ +, CD3 ~ + CD4 ~ +, CD3 ~ + CD8 ~ + and CD8 ~ + CD28 ~ + on the TIL surface of gastric cancer was lower than that in the peripheral blood and gastric cancer of normal healthy volunteers (P <0.05). The expression of CD8 ~ + CD28 ~ + on the TIL surface of gastric cancer was related to the depth of tumor invasion and tumor size (P <0.05). Conclusions The immunosuppressive status of TIL in gastric cancer may be related to the low expression of CD8 + CD28 +. The expression of CD8 + CD28 + on TIL may be used as a reference for prognosis of gastric cancer.
