目的通过检测Caspase活性分析si RNA对宫颈癌CaSki细胞凋亡的影响,评价si RNA的生物学效应。方法应用FuGene6为转染试剂,将si RNA转染入CaSki细胞,采用Western blotting检测细胞提取物中HPV16-E6的表达;用Caspase-3,-8,-9Colori metric Assay Kit试剂盒检测Caspase活性。结果si RNA抑制HPV16-E6表达;可活化Caspase-3,-8和-9。结论检测Caspase活性,能评价si RNA对宫颈癌CaSki细胞凋亡的影响。
Objective To evaluate the biological effects of si RNA by analyzing the effect of si RNA on apoptosis of cervical CaSki cells by detecting Caspase activity. Methods FuGene6 was used as a transfection reagent to transfect si RNA into CaSki cells. The expression of HPV16-E6 was detected by Western blotting. The Caspase-3, -8, -9Colori metric Assay kit was used to detect Caspase activity. Results si RNA inhibits HPV16-E6 expression; activates Caspase-3, -8 and -9. Conclusion Caspase activity can be assessed to evaluate the effect of si RNA on apoptosis of cervical cancer cells.
