为研究p16抑癌基因表达与滋养细胞癌变的关系,用免疫组化SP法测定了p16、增殖细胞核抗原(PCNA)在恶性滋养细胞肿瘤、葡萄胎及正常绒毛中的表达。结果示:①p16蛋白表达在恶性滋养细胞肿瘤和正常绒毛间差异有显著性(P<005);②p16阴性标本较p16阳性标本的PCNA阳性率显著高(P<005);③不同临床分期的恶性滋养细胞肿瘤患者p16阳性率不同,其中p16阳性患者3年生存率高于p16阴性者。以上结果提示,p16基因的突变可能是滋养细胞癌变及增殖失控的重要原因;临床检测p16蛋白表达对估计恶性滋养细胞肿瘤患者的预后有帮助。
To investigate the relationship between p16 tumor suppressor gene expression and trophoblastic carcinogenesis, the expression of p16 and proliferating cell nuclear antigen (PCNA) in malignant trophoblastic tumor, hydatidiform mole, and normal villi was measured by immunohistochemical SP method. The results showed that: 1p16 protein expression was significantly different between malignant trophoblastic tumors and normal villi (P<0. 05); 2p16 negative samples had significantly higher PCNA positive rates than p16 positive samples (P<0. 05); 3 different. The positive rate of p16 in patients with malignant trophoblastic tumors at clinical stage was different. The 3-year survival rate of p16-positive patients was higher than that of p16-negative patients. These results suggest that the mutation of p16 gene may be an important cause of trophoblastic canceration and uncontrolled proliferation. Clinical detection of p16 protein expression is helpful to estimate the prognosis of patients with malignant trophoblastic tumor.
