目的 :通过建立猪冠状动脉球囊损伤模型 ,观察普罗布考对血管内膜增生及管壁原癌基因c myc表达的影响。方法 :选用雌性NewYorkshire良种猪 19头 ,随机分为 2组 ,对照组 11头 ,普罗布考组 8头 ,均行冠状动脉球囊扩张术 ,术后喂饲 6周 ,重复冠状动脉造影后处死动物。普罗布考组于术前 4周给予普罗布考 1g ,bid ,至实验结束。常规病理制片 ,观察形态学变化 ,采用RT PCR的方法检测c mycmRNA的表达强度。 结果 :对照组损伤动脉内膜可见大量平滑肌细胞增生 ,呈偏心性增厚 ,普罗布考组内膜增厚程度明显减低。对照组c mycmRNA相对表达强度为 (2 .73± 0 .5 1) ,明显高于正常动脉 [(0 .82± 0 .0 9) ,P <0 .0 5 ],普罗布考组c mycmRNA相对表达强度为 (1.44± 0 .2 5 ) ,较对照组减弱 (P <0 .0 5 ) ,但仍高于正常动脉 (P <0 .0 5 )。结论 :普罗布考具有抑制冠状动脉球囊损伤后c myc表达和内膜增生的作用
OBJECTIVE : To observe the effect of probucol on the intimal hyperplasia and the expression of proto-oncogene c myc in porcine coronary arteries by establishing a balloon injury model of porcine coronary arteries. Methods: Thirteen female New Yorkshire breeders were randomly divided into two groups: 11 in the control group and 8 in the probucol group. All patients underwent coronary balloon dilation. After 6 weeks of postoperative feeding, they were sacrificed after coronary angiography. animal. The probucol group was given probucol 1g 4 weeks before surgery and bid was completed until the end of the experiment. Routine pathology, observed morphological changes, using RT PCR method to detect the expression of c myc mRNA. RESULTS: A large number of smooth muscle cells in the intima of the injured arteries in the control group showed hyperplasia and thickening, and the degree of intimal thickening in the probucol group was significantly reduced. The relative expression intensity of c myc mRNA in the control group was (2.73±0.51), which was significantly higher than that in the normal artery [(0.82±0.09), P<0.05]. The c myc mRNA was detected in the probucol group. The relative expression intensity was (1.44±0.55), which was lower than that of the control group (P<0.05) but still higher than that of the normal artery (P<0.05). Conclusion :Probucol can inhibit the expression of c myc and intimal hyperplasia after coronary balloon injury
