目的:通过体外实验探讨雌激素、孕激素及米非司酮对人子宫内膜癌细胞孕激素受体亚型表达的调节。方法:体外培养表达不同孕激素受体亚型的人子宫内膜癌细胞HECCA-A(PRA+)、HECCA-B(PRB+)及HECCA-AB(PRA+和PRB+),分别加入雌激素、孕激素和米非司酮,作用24、48h后采用蛋白质印迹法测定各组细胞中2种孕激素受体亚型蛋白的表达。结果:雌激素作用48 h后,HECCA-A、HECCA-AB细胞的PRA表达水平明显升高,PRA/PRB比值升高,P<0.05;孕激素及米非司酮作用48 h后,HECCA-B、HECCA-AB细胞的PRB表达水平明显降低,PRA/PRB比值升高,P<0.05。结论:雌激素上调子宫内膜癌细胞PRA的蛋白表达水平,孕激素及米非司酮下调PRB的蛋白表达水平。
OBJECTIVE: To investigate the regulation of progesterone receptor subtypes on human endometrial carcinoma cells by estrogen, progestin and mifepristone in vitro. Methods: Human endometrial carcinoma cells HECCA-A (PRA +), HECCA-B (PRB +) and HECCA-AB (PRA + and PRB +) were cultured in vitro. Estrogen, progesterone and Mifepristone, the role of 24,48 h after the Western blotting method was used to determine the expression of two kinds of progesterone receptor subtypes in each group of cells. Results: After 48 h of estrogen treatment, the expression of PRA in HECCA-A and HECCA-AB cells was significantly increased and the ratio of PRA / PRB was increased (P <0.05). After 48 h of progesterone and mifepristone treatment, B, PRB expression of HECCA-AB cells was significantly reduced, PRA / PRB ratio increased, P <0.05. CONCLUSION: Estrogen up-regulates the protein expression of PRA, progesterone and mifepristone down-regulate the protein expression of PRB.
