目的观察汉防己甲素、川芎嗪和苦杏仁甙3种中药成分对人肾成纤维细胞(KFB)的影响。方法采用ELISA法、四甲基偶氮唑蓝(MTT)法、流式细胞仪、免疫组织化学法分别检测汉防己甲素、川芎嗪、苦杏仁甙对人KFB分泌的Ⅰ型胶原酶活性、人KFB增殖、凋亡、Ⅰ型胶原表达的影响。结果汉防己甲素、川芎嗪、苦杏仁甙3种中药提取成分在各自的最佳浓度范围和作用时间内均能提高人胎KFB分泌的Ⅰ型胶原酶活性、抑制人胎KFB增殖和Ⅰ型胶原的表达、促进人胎KFB凋亡。结论汉防己甲素、川芎嗪 苦杏仁甙在预防及逆转肾间质纤维化中起重要作用。
Objective To observe the effect of three traditional Chinese medicine ingredients of tetrandrine, ligustrazine and amygdalin on human renal fibroblasts (KFB). Methods The type Ⅰ collagenase activity of human KFB secreted by tetrandrine, tetramethylpyrazine and amygdalin were detected by ELISA, MTT assay, flow cytometry and immunohistochemistry. Effects of human KFB on proliferation, apoptosis and type Ⅰ collagen expression. Results Tetrandrine, ligustrazine and amygdalin could improve the activity of type Ⅰ collagenase secreted by human fetal KFB and inhibit the proliferation of human fetal KFB and type Ⅰ Collagen expression promotes human KFB apoptosis. Conclusion Tetrandrine, ligustrazine, amygdalin play an important role in preventing and reversing renal interstitial fibrosis.
