目的:探讨通过体内阻断c-kit信号途径来建立豚鼠膀胱Cajal样细胞缺失模型的方法。方法:选取新生豚鼠20只,随机分成实验组、对照组。实验组在出生24h内的豚鼠腹腔内隔天注射c-kit抗体(ACK2)100μg,共5次,对照组腹腔内注射生理盐水,于第10天取膀胱。然后将两组豚鼠膀胱制作冰冻切片行免疫荧光染色及激光共聚焦显微镜观察。结果:10d后实验组与对照组豚鼠逼尿肌中Cajal样细胞在激光共聚焦显微镜下比较:实验组膀胱逼尿肌中Cajal样细胞数量减少甚至消失(P<0.01),Cajal样细胞二聚体消失(P<0.01)。结论:通过阻断c-kit信号通路可以成功建立豚鼠膀胱Cajal样细胞缺失模型。
OBJECTIVE: To explore a method to establish Cajal-like cell deletion model in guinea pig bladder by blocking c-kit signal pathway in vivo. Methods: Twenty newborn guinea pigs were randomly divided into experimental group and control group. The guinea pigs in the experimental group were injected intraperitoneally with c-kit antibody (ACK2) 100 μg intraperitoneally every other day for 5 times. The control group was injected intraperitoneally with saline and the bladder was taken on the 10th day. Then two groups of guinea pigs were made into frozen sections for immunofluorescence staining and laser confocal microscopy. RESULTS: After 10 days, Cajal-like cells in detrusor muscle of experimental group and control group were compared under confocal laser scanning microscope. The number of Cajal-like cells in the detrusor of experimental group decreased or even disappeared (P <0.01) Body disappeared (P <0.01). Conclusion: Cajal-like cell deletion model of bladder in guinea pigs can be successfully established by blocking c-kit signaling pathway.
