理想的药物不但具有专一性和药理活性,而且还能充分到达靶部位发挥作用。随着生物技术的发展,多肽作为药物在临床上的应用越来越广泛,至今,美国己经开发生物技术药物达300多个。临床上常用的剂型为注射溶液剂和冻干粉针剂。由于此类药物在体内极易降解,半衰期较短,必须频繁给药,这给患者带来诸多的不便和加重其经济负担。因此,相应的制剂学、给药方式以及药物在体内的转运的途径日益受到医学各界的关注。国外自80年代开始主要从非注射途径给药及长效注射剂两方面来研究新一代剂型。本文着重从给药方式及其在体内循环的途径加以综述。
Ideal drugs not only have specificity and pharmacological activity, but also fully reach the target site to play a role. With the development of biotechnology, polypeptides are increasingly used clinically as medicines. To date, more than 300 biotech drugs have been developed in the United States. Commonly used in clinical dosage form injection solution and freeze-dried powder. Because these drugs easily degrade in the body, short half-life, must be frequently administered, which brings a lot of inconvenience to patients and increase their financial burden.
