目的研究中药骨乐冲剂对体外培养大鼠成骨细胞分化和增殖的影响,探讨其调节机制。方法采用双酶消化法获得大鼠成骨细胞,传代培养3代后用3种浓度的骨乐冲剂含药血清培养。采用MTT法测定成骨细胞增殖率和碱性磷酸酶活性。结果含高、中浓度骨乐冲剂的血清能显著促进成骨细胞的增殖并提高碱性磷酸酶活性。结论中药骨乐冲剂能够促进成骨细胞的增殖与分化。
Objective To study the effect of Gualu granule on differentiation and proliferation of rat osteoblasts cultured in vitro and its regulation mechanism. Methods The rat osteoblasts were obtained by digestion with double enzymes. After subculture for 3 generations, the osteoblasts were cultured with three concentrations of Gulei granules. The proliferation rate of osteoblasts and alkaline phosphatase activity were measured by MTT assay. Results Serum containing high and medium concentration Gulei granules could significantly promote the proliferation of osteoblasts and increase the alkaline phosphatase activity. Conclusion Gualule granules can promote the proliferation and differentiation of osteoblasts.
