目的 :用高效液相色谱法测定血浆中鹿蹄草素的浓度。方法 :采用YWG -C1 8色谱分析柱 (4 6mm× 2 5 0mm ,5 μm) ,WatersTM486可变波长紫外检测器 ,甲醇 -水 (4 0∶6 0 )为流动相 ,香草醛为内标。血液样品用乙醚提取纯化后进样 ,在2 2 0nm波长处检测。结果 :鹿蹄草素线性范围为 1 0~ 10 0 0mg·L- 1 (r=0 995 3) ,最低检测量 0 1μg ,最低检出浓度 5 0mg·L- 1 ,平均方法回收率 98 36 % ,精密度考察鹿蹄草素日内、日间RSD均小于 4%。结论 :本方法专属性高 ,快速 ,准确 ,重复性好。
Objective: To determine the concentration of salidroside in plasma by high performance liquid chromatography. Methods: YWG-C8 chromatographic column (4.6 mm × 250 mm, 5 μm), Waters ™ 486 variable wavelength UV detector with methanol-water (40:60) as mobile phase and vanillin as internal standard. The blood samples were extracted with ether and purified for injection, and detected at a wavelength of 220 nm. Results: The linear range of gentiana was 10 ~ 100 mg · L -1 (r = 0 995 3), the lowest detection limit was 0 1 μg and the lowest detection limit was 50 mg · L -1. The average recoveries were 98 36 %, Precision test of Pyrenarrhea day, day RSD were less than 4%. Conclusion: The method is highly specific, fast, accurate and reproducible.
