普通小麦品种中国春小麦(Chinese spring)及其突变体中国春ph与新疆大赖草(Legmus racomosus(Lam.)Tzvel)杂交授粉后12～13天的400～600μm大小的幼胚来用一步成苗法(幼胚→幼苗)和二步成苗法(幼胚→愈伤组织→幼苗)进行离体培养.在Ms基本培养基+3～7%蔗糖+400毫克/升乳蛋白水解物或Norstog’s氨基酸组分的培养基中培养的幼胚,平均成苗率为65.8%,添加5%蔗糖和Norstog’s氨基酸组分的培养基中成苗率最高80%.而小于400微米的幼胚仅能在附加7%蔗糖和Norstog’s氨基酸组分的培养基中萌发成苗.将幼胚接种在Ms+2,4-D2毫克/升+天冬素150毫克/升+蔗糖2%的培养基中,诱导形成愈伤组织,愈块在Ms+IAA1毫克/升+6-BA1毫克/升+蔗糖2%的培养基中再生出绿苗.9个月内内中国春/新疆大赖草单胚成苗23.5株;中国春(ph)/新疆大赖草为12.5株.
