目的:证实鬼臼毒素自旋标记物GP4和GP7体外对Molt4B细胞周期和大分子合成的抑制作用.方法:MTT、同位素标记法和流式细胞分光光度仪BrdU/DNA分析法.结果:GP4,GP7和依托泊苷(002-100mmol·L-1,48h)抑制细胞生长量效曲线不同,其IC50值分别为0.11,4.7,1.6mmol·L-1,并降低DNA、蛋白质合成.GP4(10mmol·L-112h)增加有丝分裂指数,为对照组的5倍;GP7和依托泊苷都降低有丝分裂指数,分别为对照组的1/3和1/10.GP4主要阻止细胞于G2/M期,而GP7与依托泊苷相似,将细胞阻滞在S和G2期.结论:GP4和GP7抑制Molt4B细胞生长和大分子合成,其作用机制不同
OBJECTIVE: To demonstrate the inhibitory effect of podophyllotoxin spin markers GP4 and GP7 on Molt4B cell cycle and macromolecular synthesis.
