目的:研究重组人白细胞介素6(rhIL-6)对原代培养大鼠肝细胞分泌纤维蛋白原(Fibrinogen,Fgn)的影响及蛋白激酶C(PKC)抑制剂Ro31-8220对枯否细胞分泌IL-6的作用。方法:以酶联免疫吸附测定法(ELISA)测定Fgn;测定I-6采用[3H]-TdR掺入增殖法。结果:大鼠肝细胞(3×1O6/ml)与rhIL-6(0~100kU/L)孵育24h,刺激肝细胞分泌Fgn增加;Ro31-8220(0.1~10μmol/L)显著抑制枯否细胞释放IL-6。结论:提示IL-6是调节纤维蛋白原合成的重要细胞因子,推测PKC抑制剂可通过抑制枯否细胞分泌IL-6来降低Fgn的分泌。
AIM: To investigate the effect of recombinant human interleukin-6 (IL-6) on the fibrinogen (Fgn) secretion of primary cultured rat hepatocytes and the effect of protein kinase C (PKC) inhibitor Ro31-8220 on the secretion of Kupffer cells The role of IL-6. Methods: Enzyme-linked immunosorbent assay (ELISA) was used to determine Fgn. I-6 was assayed by [3H] -TdR incorporation assay. Results: After incubated with 3 × 10 6 / ml rhIL-6 (0-100 kU / L) for 24 hours, the hepatocytes secreted Fgn increased. Ro31-8220 (0.1-10 μmol / L) Cells release IL-6. CONCLUSION: It is suggested that IL-6 is an important cytokine that regulates fibrinogen synthesis. It is speculated that PKC inhibitor can decrease Fgn secretion by inhibiting the secretion of IL-6 from Kupffer cells.
