目的探讨了血管舒缓素对血小板趋化生长因子(PDGF)诱导培养的大鼠胸主动脉平滑肌细胞增殖的影响。方法取SD大鼠胸主动脉平滑肌细胞进行培养,传至5代后,分组观察PDGF、PDGF+血管舒缓素组、PDGF+血管舒缓素+缓激肽受体拮抗剂组等。干预因素作用于血管平滑肌细胞(VSMC),对3H胸腺嘧啶(3HTdR)的掺入及原癌基因cmycmRNA表达的影响。结果1.PDGF可促进VSMC3HTdR的掺入和cmycmRNA表达,这种表达在2小时时达高峰。2.血管舒缓素可抑制PDGF诱导的VSMC3HTdR的掺入和cmycmRNA表达,这种抑制作用可被缓激肽受体拮抗剂所阻断。结论血管舒缓素可抑制PDGF诱导的VSMC增殖作用。
Objective To investigate the effects of kallikrein on the proliferation of thoracic aortic smooth muscle cells induced by platelet chemotactic growth factor (PDGF) in rats. Methods Thoracic aorta smooth muscle cells from SD rats were cultured and passaged for 5 generations. PDGF, PDGF + kallikrein group, PDGF + kallikrein + bradykinin receptor antagonist group were observed. The effect of intervention on vascular smooth muscle cells (VSMCs), 3H-thymidine incorporation (3H-TdR) and oncogene c-mycmRNA expression. Results 1. PDGF can promote VSMC3H TdR incorporation and c mycmRNA expression, this expression reached its peak at 2 hours. 2. Vasopressin can inhibit PDGF induced VSMC3H TdR incorporation and c mycmRNA expression, this inhibition can be blocked by bradykinin receptor antagonists. Conclusions Vasopressin can inhibit PDGF-induced VSMC proliferation.
