目的研究地锦草提取物对红色毛癣菌角鲨烯环氧化酶和β-(1,3)-D-葡聚糖合成酶的影响,探讨其抗真菌作用机制。方法药物干预红色毛癣菌后分离菌体,按试剂盒要求处理样品,利用酶联免疫法和荧光分光光度法,研究地锦草提取物对红色毛癣菌角鲨烯环氧化酶和β-(1,3)-D-葡聚糖合成酶活性的作用。结果地锦草提取物256μg.m l-1时,可显著降低红色毛癣菌中角鲨烯环氧化酶的活性(P<0.01);不同浓度的地锦草提取物,均可降低红色毛癣菌β-(1,3)-D-葡聚糖合成酶的活性(P<0.01)。结论地锦草提取物抗真菌机制可能与抑制角鲨烯环氧化酶和β-(1,3)-D-葡聚糖合成酶的活性有关。
Aim To study the effect of extract of Eucheuma on Trichophyton rubrum squalene cyclooxygenase and β- (1,3) -D-glucan synthase and to explore its antifungal mechanism. Methods The drug was isolated from Trichophyton rubrum, and then the samples were processed according to the requirements of the kit. The enzyme-linked immunosorbent assay (FTIR) and fluorescence spectrophotometry were used to study the effects of extract of Eucheuma on Trichophyton rubrum squalene epoxidase and beta - (1,3) -D-glucan synthase activity. Results The extract of Magnolia bark was significantly inhibited by squalene epidermidis activity (P <0.01) at the concentration of 256 μg.m l-1. The extract of Eucheuma at different concentrations reduced red Trichophyton rubrum β- (1,3) -D-glucan synthase activity (P <0.01). Conclusion The antifungal mechanism of extracts of Eucheuma extract may be related to the inhibition of the activities of squalene cyclooxygenase and β-1,3-D-glucan synthase.
