目的:观察通络止痛方对人膝骨关节炎滑膜炎性细胞TLR4、IL-1β及NO的影响。方法:取本院行全膝关节置换术患者术中废弃滑膜组织培养成纤维细胞进行不同浓度浓度通络止痛方干预,具体分为:高浓度组(2.5mg/m L)、中浓度(0.25mg/m L)、低浓度组(0.025mg/m L)、对照组。应用PCR检测细胞内TLR4 m RNA表达情况,采用ELISA检测细胞培养上清液中IL-1β、NO浓度的变化。结果:高、中、低浓度组较对照组上清液中IL-1β及NO含量显著降低(P<0.05),且下降的幅度与通络止痛方的浓度呈正相关(1L-1β:r=0.39,P<0.05;NO:r=0.52,P<0.05)。与对照组比较,高、中浓度组TLR4 m RNA的表达水平显著降低(P<0.05)。低浓度组TLR4 m RNA抑制作用与对照组差异无统计学意义。中、高浓度组抑制作用与浓度呈正比。结论:提示通络止痛方可能通过Toll样受体通路影响或抑制人膝骨关节炎滑膜炎性反应。
Objective: To observe the effect of Tongluzhitong Recipe on TLR4, IL-1β and NO in synovial inflammatory cells of human knee osteoarthritis. Methods: The total knee arthroplasty in patients with intraoperative synovial tissue culture of fibroblasts for different concentrations of Tongluzhitong Fang intervention, divided into: high concentration group (2.5mg / m L), the concentration ( 0.25mg / m L), low concentration group (0.025mg / m L), control group. The expression of TLR4 m RNA in cells was detected by PCR, and the concentration of IL-1β and NO in cell culture supernatants was detected by ELISA. Results: The levels of IL-1β and NO in the supernatant of the high, medium and low concentration groups were significantly lower than those in the control group (P <0.05), and the decreasing amplitude was positively correlated with the concentration of Tongluzhitongfang (1L-1β: r = 0.39, P <0.05; NO: r = 0.52, P <0.05). Compared with the control group, the expression levels of TLR4 mRNA in high and middle concentration groups were significantly decreased (P <0.05). The inhibitory effect of TLR4 m RNA in low concentration group was not significantly different from that in control group. Medium and high concentrations of inhibition was proportional to the concentration. Conclusion: It suggests that Tongluzhitong may affect or inhibit synovitis of human knee through Toll-like receptor pathway.
