目的:探讨中药健乳灵对DMBA诱导乳腺不典型增生大鼠乳腺组织学及血清性激素的影响。方法:6周龄SPF级雌性SD大鼠32只,随机分为正常组、模型组、健乳灵组、他莫昔芬组,模型组、健乳灵组、他莫昔芬组行DMBA灌胃造模,正常组和模型组给于常规饲料,健乳灵组和他莫昔芬组分别给于健乳灵饲料、他莫昔芬饲料喂养70天。观察各组大鼠乳腺组织学改变,比较各组大鼠血清雌二醇、孕酮、睾酮水平。结果:模型组乳腺组织呈现不典型增生改变,健乳灵组、他莫昔芬组乳腺组织轻度增生;与正常组比较,模型组雌二醇、孕酮水平明显升高,睾酮明显降低,他莫昔芬组雌二醇、孕酮、睾酮水平明显下调;健乳灵组雌二醇、孕酮水平降低,睾酮水平与正常组接近。结论:健乳灵可改善DMBA所致大鼠乳腺不典型增生,其机制与调节大鼠性激素水平紊乱有关。
Objective: To investigate the effect of Jiankailing on breast histology and serum sex hormones in DMBA-induced mammary dysplasia rats. Methods: Thirty-two SPF female Sprague-Dawley rats of 6 weeks old were randomly divided into normal group, model group, Jiankailing group, tamoxifen group, model group, Jiankailing group and tamoxifen group. Gastric modeling, normal group and model group were given regular diet, Jiankailing group and tamoxifen group were given Jiankuling feed, tamoxifen feed for 70 days. The histological changes of mammary gland in each group were observed. The levels of serum estradiol, progesterone and testosterone were compared between the two groups. Results: The model group showed atypical hyperplasia of mammary gland tissues. The breast tissues of JKL group and tamoxifen group were mildly hyperplastic. Compared with the normal group, the levels of estradiol and progesterone were significantly increased and the testosterone levels were significantly decreased The levels of estradiol, progesterone and testosterone in tamoxifen group were significantly decreased. The levels of estradiol and progesterone in Jiank Lingling group were lower than those in normal group. Conclusion: Jiankailing can improve atypical hyperplasia of mammary gland induced by DMBA in rats, and its mechanism is related to the disorder of sex hormone in rats.
