目的 探讨癌细胞体外经抗癌药作用后残留的端粒酶活性作为判断化疗敏感性的可能性。方法 用噻唑兰(MTT)法和端粒酶活性法(TA)同时测定17例大肠癌对5-氟尿嘧啶、丝裂霉素、顺氯氨铂的体外敏感性。结果 有1例肠癌当细胞数为8×10~2时,MTT法和端粒酶活性法测其OD值分别为接近0和0.364,又有1例肠癌MTT法测得对照组OD值仅0.039,用药组接近0,但用端粒酶活性法测得OD值为0.378,5-Fu组为0.32,DDP组为0.153,MMC组为0.02。上述事实说明端粒酶活性法灵敏度远高于MTT法。另外,用端粒酶活性法测得抗癌药抑制率大于MTT法,其中DDP组存在显著差异。结论 端粒酶活性法可以作为一种新的体外判断抗癌药敏感性的方法。
Objective To investigate the possibility of telomerase activity remaining after cancer cells are treated with anticancer drugs in vitro as a possibility to determine the sensitivity of chemotherapy. Methods The in vitro sensitivity of 17 cases of colorectal cancer to 5-fluorouracil, mitomycin and cisplatin were determined by thiazolyl (MTT) method and telomerase activity method (TA). Results In 1 case of colorectal cancer, when the cell number was 8×10~2, the OD values measured by MTT assay and telomerase activity method were close to 0 and 0.364, respectively, and 1 case of colon cancer was measured by MTT method to determine the OD value of the control group. Only 0.039, the drug group was close to 0, but the OD value was 0.378 by the telomerase activity assay, 0.32 in the 5-Fu group, 0.153 in the DDP group, and 0.02 in the MMC group. The above facts indicate that the sensitivity of telomerase activity method is much higher than that of MTT method. In addition, the inhibitory rate of anticancer drugs was greater than that of MTT assay using telomerase activity assay, among which there was a significant difference in DDP group. Conclusion Telomerase activity assay can be used as a new method to judge the sensitivity of anticancer drugs in vitro.
