目的观察染尘巨噬细胞上清液对人胚肺成纤维细胞热休克蛋白70(HSP70)各亚型(HSP70-1、HSP70-2、HSP70-hom)表达的影响。方法采用大鼠巨噬细胞(AMs)和人胚肺成纤维细胞(HELF)构成体外模型,采用逆转录多聚酶链反应(RT-PCR)技术检测二氧化硅粉尘处理AMs24h的培养上清液刺激HELF细胞的HSP70各亚型表达情况。同时用二氧化钛作为阴性对照,以不用粉尘处理的AM培养上清液作为正常对照。结果HSP70-1、HSP70-2、HSP70-hom基因表达与粉尘浓度的相关系数(R)依次为0.931(P<0.01)、0.923(P<0.01)、0.818(P<0.05),呈现明显的剂量-反应关系。结论受染尘肺泡巨噬细胞上清液刺激的人胚肺成纤维细胞HSP70各亚型在转录水平呈表达增加,且可能参与了矽尘致细胞氧化应激过程。
Objective To observe the effects of the supernatant of the macrophages on the expression of heat shock protein 70 (HSP70) subtypes (HSP70-1, HSP70-2, HSP70-hom) in human embryo lung fibroblasts. Methods Rat macrophages (AMs) and human embryonic lung fibroblasts (HELF) were used to construct in vitro models. The culture supernatants of AMs24h treated with silica dust were detected by reverse transcription polymerase chain reaction (RT-PCR) HSP70 subtypes of cells expression. At the same time, titanium dioxide was used as a negative control, and dust-free AM culture supernatant was used as a normal control. Results The correlation coefficients (R) between HSP70-1, HSP70-2 and HSP70-hom gene expression and dust concentration were 0.931 (P <0.01), 0.923 (P <0.01) and 0.818 (P < - Response relationship. Conclusions The HSP70 isoforms of human embryonic lung fibroblasts stimulated by the pneumocytes are expressed at transcriptional level and may be involved in the oxidative stress induced by silica dust.
