目的 :探讨滑膜素(synoviolin)在乳腺癌组织中的表达情况及其对乳腺癌细胞MCF-7增殖和迁移能力的影响。方法 :通过Western blot、免疫组织化学染色及免疫组织芯片技术检测乳腺癌组织中滑膜素的表达水平;在乳腺癌细胞MCF-7中利用腺病毒过表达滑膜素,并以划痕实验、Transwell迁移实验、CCK-8细胞增殖试剂盒等检测其对于乳腺癌细胞MCF-7迁移和增殖能力的影响。结果:滑膜素在乳腺癌组织中的表达水平明显低于相应癌旁组织;过表达滑膜素能抑制乳腺癌细胞MCF-7的增殖和迁移能力;过表达滑膜素能抑制乳腺癌细胞MCF-7发生上皮-间质转化。结论:滑膜素在乳腺癌组织中低表达,可能与抑制乳腺癌细胞的迁移和增殖能力密切相关。
Objective: To investigate the expression of synoviolin in breast cancer and its effect on the proliferation and migration of breast cancer cell line MCF-7. Methods: The expression of synoviolin in breast cancer tissues was detected by Western blot, immunohistochemistry and immunohistochemistry. The adenovirus overexpression of synovin was used in breast cancer cells MCF-7, Transwell migration assay and CCK-8 cell proliferation kit were used to detect the effect of MCF-7 on the migration and proliferation of breast cancer cells. Results: The expression of synovin in breast cancer tissues was significantly lower than that in paracancerous tissues; overexpression of synovitis could inhibit the proliferation and migration of breast cancer cells MCF-7; overexpression of synovitis could inhibit breast cancer cells Epithelial-mesenchymal transition occurs in MCF-7. Conclusions: The low expression of synoviolin in breast cancer tissues may be closely related to the inhibition of the migration and proliferation of breast cancer cells.
