目的 :探讨食管癌及癌旁组织中 p53、MDR基因的表达及其临床意义。 方法 :采用免疫组化ABC法及逆转录一聚合酶链反应的方法 ,对 50例食管癌和 37例癌旁组织中p53、MDR基因的表达情况进行检测。结果 :p53基因蛋白在食管癌中有显著表达 70 % (35/ 50 ) ,癌旁组织中有表达 62 % (2 3/37) ;其中在正常组织中占 36 % (5/ 1 4 ) ,在非典型增生中占 78% (1 8/ 2 3)。MDR基因在食管癌高、中、低分化中的表达水平有极显著意义 ,癌与正常组织中有极显著意义。在不同年龄、性别、肿瘤大小上 p53、MDR基因的表达无显著意义。结论 :p53、MDR基因在食管癌中均呈高水平表达 ,癌旁组织中MDR有表达 ,p53基因突变早于组织形态学改变
Objective: To investigate the expression of p53 and MDR genes in esophageal carcinoma and its adjacent tissues and their clinical significance. Methods: The expressions of p53 and MDR genes in 50 cases of esophageal cancer and 37 cases of paracancerous tissues were detected by immunohistochemical ABC method and reverse transcription polymerase chain reaction (RT-PCR). Results: The p53 gene protein was highly expressed in esophageal carcinoma (70%, 35/50) and adjacent tissues (62%, 3/36) respectively, which accounted for 36% (5/14) in normal tissues, In atypical hyperplasia accounted for 78% (18/23). MDR gene expression in esophageal cancer in high, moderate, poorly differentiated levels have very significant, cancer and normal tissue has a very significant meaning. In different age, gender, tumor size, p53, MDR gene expression was not significant. Conclusions: The expression of p53 and MDR genes in esophageal carcinoma was high, and the expression of MDR in adjacent tissues was also observed. The mutation of p53 gene was earlier than that in histomorphology
