目的了解白细胞介素-6(IL-6)和白细胞介素-8(IL-8)对子宫异位内膜基质细胞分泌基质金属蛋白酶-9(MMP-9)的影响,以探讨IL-6及IL-8在异位内膜粘附和种植过程中的作用。方法分别用含不同浓度的IL-6和IL-8以及无干扰因素的培养液培养各组人子宫内膜基质细胞24h后,ELISA方法测定培养液中MMP-9的含量。结果1.正常对照组的MMP-9水平:高浓度IL-6干扰组显著高于无干扰组(P<0.05)。2.在位内膜组和异位内膜组的MMP-9水平:⑴高浓度IL-6干扰组显著高于低浓度IL-6干扰组(P<0.01)。⑵高浓度IL-6干扰组显著高于无干扰组(P<0.01)。结论内异症时IL-6可能是通过产生更多MMP-9的途径来促进异位内膜种植,而IL-8促进异位内膜种植不是通过提高MMP-9水平,而是直接刺激内膜细胞增殖,从而对异位内膜组织的生长和维持起作用。
Objective To investigate the effects of interleukin-6 (IL-6) and interleukin-8 (IL-8) on the secretion of matrix metalloproteinase-9 (MMP-9) in uterine ectopic stromal cells And IL-8 in the ectopic endometrial adhesion and the role of planting. Methods Human endometrial stromal cells were cultured with culture medium containing different concentrations of IL-6 and IL-8 as well as non-interfering factors for 24 h. ELISA was used to determine the content of MMP-9 in the culture medium. MMP-9 levels in normal control group: high-concentration IL-6 interference group was significantly higher than non-interference group (P <0.05). MMP-9 levels in eutopic endometrium group and ectopic endometrium group: (1) The levels of MMP-9 in high concentration IL-6 interference group were significantly higher than those in low concentration IL-6 interference group (P <0.01). ⑵ high concentration of IL-6 interference group was significantly higher than non-interference group (P <0.01). Conclusions IL-6 may promote ectopic endometrial implantation by generating more MMP-9 during endometriosis. However, IL-8 promotes ectopic endometrial implantation not by increasing MMP-9 but directly stimulating Membrane cells proliferate, thus contributing to the growth and maintenance of ectopic endometrial tissue.
