目的观察银杏叶提取物对肺纤维化大鼠不同时间点肺组织MMP-2mRNA表达的影响。方法 96只健康Wistar大鼠随机分为正常对照组(A组)、模型组(B组)、强的松组(C组)和银杏叶提取物干预组(D组),每组各24只,除A组外,其余各组采用博莱霉素复制大鼠肺纤维化模型。造模第2天起,D组用银杏叶提取物灌胃,C组用醋酸强的松混悬液灌胃,A、B组用生理盐水灌胃。每组分别于造模后第7、14、28天随机取8只大鼠,处死后采集肺组织,抽提RNA,采用RT-PCR法得到MMP-2mRNA产物,用琼脂糖凝胶电泳和图像分析方法观察MMP-2 mRNA的表达。结果各实验组在各时间点均有MMP-2mRNA的表达,与A组相比,B组第7天含量显著升高(P<0.01),第14天时达到高峰,第28天虽较第7、14天时有所降低,但仍明显高于同期A组(P<0.01)。与B组比较,C、D组各时间点含量均明显降低(P<0.01)。结论造模后各组MMP-2mRNA表达增强,银杏叶提取物能够有效抑制MMP-2mRNA的过度表达,从而对肺损伤和肺纤维化起到一定的防治作用。
Objective To observe the effect of Ginkgo biloba extract on the expression of MMP-2 mRNA in lung tissue of rats with pulmonary fibrosis at different time points. Methods 96 healthy Wistar rats were randomly divided into normal control group (group A), model group (group B), prednisone group (group C) and ginkgo biloba extract intervention group (group D) , Except A group, the rest of the group using bleomycin replication rat lung fibrosis model. From the second day of model making, group D was gavaged with ginkgo biloba extract, group C was given gavage with prednisone acetate, and group A and group B were given gavage with saline. Eight rats were randomly selected from each group on the 7th, 14th and 28th day after the model was established. The lung tissues were harvested and RNA was extracted. The MMP-2 mRNA was obtained by RT-PCR. The agarose gel electrophoresis and image Analyze the expression of MMP-2 mRNA. Results Compared with group A, the content of MMP-2 mRNA in group B was significantly increased at the 7th day (P <0.01), and peaked at the 14th day 7, 14 days decreased, but still significantly higher than the same period A group (P <0.01). Compared with group B, the contents of C and D at each time point were significantly decreased (P <0.01). Conclusion The expression of MMP-2mRNA in each group increased after modeling, and Ginkgo biloba extract could effectively inhibit the overexpression of MMP-2mRNA, which could prevent lung injury and pulmonary fibrosis.
