测定血浆和淋巴细胞内环孢素A结合蛋白的含量。用鼠源单抗H5 作包被抗体, 并以酶标记兔抗环孢素A结合蛋白(CyP) , 建立了检测CyP的双抗体夹心ELISA, 检测了25 例健康献血员和36 例全身性红斑狼疮(SLE) 患者血浆和淋巴细胞内CyP水平。结果显示, 检测下限为50μg/L, 批内和批间变异系数分别为5-4% 和9-2% , 回收率为95-3% 。献血员血浆CyP水平为(165±80 )μg/L, 淋巴细胞内为(2290 ±380)μg/g 蛋白。血浆和淋巴细胞CyP水平无相关(r= - 0-12) 。SLE患者血浆CyP为(325±420)μg/L, 淋巴细胞内为(2640±650)μg/g 蛋白, 均高于正常对照( P< 0-05) 。活动期SLE 病人血浆和淋巴细胞CyP水平高于非活动期病人(P< 0-05) 。该方法简便、灵敏、特异, 血浆和淋巴细胞内CyP水平与SLE的活动性有一定的联系
The contents of cyclosporin A binding protein in plasma and lymphocytes were determined. The McAb H5 was used as the coating antibody, and the rabbit anti-cyclosporin A binding protein (CyP) was labeled with enzyme to establish a double-antibody sandwich ELISA for detection of CyP. Twenty-five healthy blood donors and 36 cases of systemic erythema CyP levels in the plasma and lymphocytes of patients with lupus (SLE). The results showed that the detection limit was 50μg / L, the intra-assay and inter-assay coefficient of variation were 5-4% and 9-2% respectively, and the recovery was 95-3%. Blood donors plasma CyP levels (165 ± 80) μg / L, lymphocytes (2290 ± 380) μg / g protein. Plasma and lymphocyte CyP levels were not correlated (r = - 0-12). The plasma levels of CyP in SLE patients were (325 ± 420) μg / L and (2640 ± 650) μg / g in lymphocytes, both of which were higher than those of normal controls (P <0-05). The plasma and lymphocyte CyP levels of active SLE patients were higher than inactive patients (P <0-05). The method is simple, sensitive and specific, and the level of CyP in plasma and lymphocytes is related to the activity of SLE
