为了筛选东北刺人参不定根组培增殖适宜的培养基成分,以东北刺人参不定根为外植体,研究了MS培养基中NH4+/NO3-和有机物质对东北刺人参不定根增殖生长和分化的影响,同时观察了培养过程中不定根的生长动态。结果表明:MS培养基中NH4+/NO3-为30/30mmol时适宜不定根的生长,鲜物质量和干物质量优于其他MS培养基中NH4+/NO3-,分别为809mg和117mg。当有机物质含量为MS原配方量的1.5倍时,不定根生长较好。通过观察不定根的生长动态发现,培养14d后不定根数随着培养天数的增加而增加,在培养24~26d时东北刺人参不定根生长达到旺盛期,此后不定根数随着培养天数增加而不再增加。
In order to screen the suitable culture medium components of adventitious root tissue culture of Northeast Ginseng, adventitious roots of A. ginseng were used as explants to study the effect of NH4 + / NO3- and organic matter in MS medium on the adventitious root growth and differentiation of A. japonicus adventitious roots. At the same time, the growth of adventitious roots in culture was observed. The results showed that MS medium was suitable for adventitious root growth when the NH4 + / NO3- ratio was 30 / 30mmol, and the quality of fresh and dry matter was better than that of NH4 + / NO3- in other MS mediums, which were 809mg and 117mg, respectively. When the content of organic matter is 1.5 times the original amount of MS, adventitious root growth is better. By observing the dynamic growth of adventitious roots, we found that the number of adventitious roots increased with the number of culture days after 14 days of culture. The adventitious root growth of A. japonicus reached to the peak during 24-26 days after culture, and the number of adventitious roots did not increase with the increase of days after culturing.
