目的通过对糖类成分的两种色谱方法进行比较,选择一种准确、方便、快速测定注射用丹参(冻干)中糖类成分的方法。方法采用高效液相-蒸发光散射法(HPLC-ELSD),Alltech PrevailTMCarbohydrate ES(4.6 mm×250 mm,5μm)色谱柱,乙腈-水(76∶24)流动相等度洗脱,ELSD检测器。采用离子色谱法(IC),Carbo Pac PA20(3 mm×150 mm,S/N 003654)阴离子交换分析色谱柱,Carbo Pac PA20(3 mm×50 mm,S/N 003405)保护柱,以氢氧化钠溶液(7 mmol/L)洗脱,脉冲安培检测器。以上两种色谱方法分别对注射用丹参(冻干)中糖类成分进行测定。结果在上述色谱条件下,HPLC-ELSD法可同时测定果糖和葡萄糖2种糖类成分,方法的线性关系、精密度、重复性及回收率等均符合要求;IC法可同时测定阿拉伯糖、半乳糖、葡萄糖、蔗糖、甘露糖和果糖6种糖类成分,方法经考察准确可靠。两种色谱方法分别测定的果糖和葡萄糖结果基本一致。结论离子色谱法更适合于注射用丹参(冻干)中糖类成分的测定。
OBJECTIVE To compare the two chromatographic methods of carbohydrate components and select a method for accurate, convenient and rapid determination of carbohydrate components in Salvia miltiorrhiza for injection. Methods HPLC-ELSD, Alltech PrevailTM Carbohydrate ES (4.6 mm × 250 mm, 5 μm) column was used to elute the mobile phase of acetonitrile-water (76:24). The ELSD detector was used. The column was analyzed by ion chromatography (IC), Carbo Pac PA20 (3 mm × 150 mm, S / N 003654) anion exchange chromatography column and Carbo Pac PA20 (3 mm × 50 mm, S / N 003405) Sodium solution (7 mmol / L) elution, pulsed amperometric detector. The two chromatographic methods were used for injection of Salvia (freeze-dried) sugar content was determined. Results Under the above chromatographic conditions, the HPLC-ELSD method was able to simultaneously determine the two saccharide components of fructose and glucose. The linearity, precision, reproducibility and recovery of the method met the requirements. The IC method can simultaneously determine arabinose, Lactose, glucose, sucrose, mannose and fructose six kinds of sugar composition, the method is accurate and reliable after inspection. Fructose and glucose respectively determined by the two chromatographic methods were basically the same. Conclusion Ion chromatography is more suitable for the determination of carbohydrate components in Salvia miltiorrhiza for injection.
