目的:探讨补肾法中药复方干预性治疗的作用机理,为中医药调节女性生殖轴的研究提供一个新的符合中医证候特点的病证结合动物模型。方法:选雌性育龄期SD大鼠行阴道脱落细胞学涂片,筛选出动情周期正常者为受试动物,除正常组外,其余大鼠予雷公藤多苷致动情周期紊乱模型。雷公藤模型大鼠随机分为即空白组、倍美力组、中药高、中、低剂量组5组,分别予倍美力和通脉大生片不同剂量治疗7天,空白组同期给予等量生理盐水,正常组正常喂养。各组均于治疗前后采血,称重法测算卵巢指数;放射免疫法检测血清雌二醇、孕酮;酶联免疫吸附法检测抑制素B水平;免疫组化法检测卵巢雌激素受体(ER)、孕酮受体(PR)。结果:①予雷公藤多苷40 mg/kg.d连续灌胃10周,可造成育龄期大鼠动情周期紊乱或消失,卵巢指数下降,血中E2、INHB水平明显降低;卵巢组织中ER表达明显降低;②运用通脉大生片干预性治疗后,大鼠动情周期恢复至正常范围,卵巢指数升高;血清E2、INHB水平及卵巢组织中ER表达均明显升高;与空白组相比均有显著统计学意义。结论:雷公藤多苷可造成大鼠卵泡发育障碍和卵巢功能低下,补肾气益精血的中药复方“通脉大生片”能显著提高模型大鼠血清雌二醇和抑制素B水平及卵巢中ER表达,从而有效地提高雌激素的生物效应和卵巢储备功能。
OBJECTIVE: To explore the mechanism of TCM intervention by tonifying kidney and to provide a new animal model of disease-syndrome combined with traditional Chinese medicine syndromes for the study of traditional Chinese medicine regulating female reproductive axis. Results: ① Tripterygium wilfordii glycosides 40 mg / kg.d for 10 weeks, can cause disorder or disappearance of estrous cycle in ovariectomized rats, decreased ovarian index, E2, INHB levels in blood significantly decreased; ER expression in ovarian tissue Significantly decreased; ②Through Tongmaisashengdan intervention treatment, the estrous cycle of rats returned to normal range, ovarian index increased; serum E2, INHB levels and ovarian tissue ER expression were significantly increased; compared with the blank group There is significant statistical significance. Conclusion: Tripterygium glycosides can cause obstructive follicular development and ovarian dysfunction in rats, and the traditional Chinese medicine compound “Tongmaishengsheng Tablet” of Bushen Qi Yi Jingxue can significantly increase serum estradiol and inhibin B levels and ovarian In ER expression, which effectively improve the biological effects of estrogen and ovarian reserve.
