目的 :研究用妊娠糖尿病 ( GDM)患病同胞对 DNA验证 2型糖尿病的易感基因 ,并找寻与 GDM连锁的易感基因。方法 :采用患病同胞对分析法 ,通过连锁分析发现易感位点。所选用的微卫星标记均是被报道已与 2型糖尿病连锁。结果 :在对 1 0 1对妊娠糖尿病患病同胞对进行连锁分析结果显示 :D1 0 S5 87为连锁位点。( NPL Score=1 .85 5 ,P=0 .0 4 7;Mean IBD=0 .5 8,P=0 .0 2 5 6)。结论 :具有 2型糖尿病致病基因的 1 0 q2 6区域同样具有致GDM的基因。一旦致病基因被定位 ,可为糖尿病的预防、诊断、治疗和药物开发提供重要的理论基础和指导。
OBJECTIVE: To study susceptibility genes of DNA for type 2 diabetes mellitus (T2DM) in pregnant women with gestational diabetes mellitus (GDM) and to find susceptibility genes linked to GDM. Methods: Using sibling pair analysis, the susceptibility loci were found by linkage analysis. The selected microsatellite markers are reported to be linked to type 2 diabetes. RESULTS: A linkage analysis of the diseased sibs of the 110 pairs of gestational diabetes showed that D1 0 S5 87 was a linkage site. (NPL Score = 1 .85 5, P = 0 .0 4 7; Mean IBD = 0 .5 8, P = 0 .0256). CONCLUSION: The 10 q26 region with pathogenic type 2 diabetes also has GDM-induced genes. Once the pathogenic genes are located, it can provide important theoretical basis and guidance for the prevention, diagnosis, treatment and drug development of diabetes.
