采用细胞毒试验、肿瘤细胞杀伤试验、集落形成试验和生长抑制试验等多种方法,检测了东亚钳蝎Buthus martensil karsch蝎毒组分Ⅱ(SVC Ⅱ)对人喉痛HEp-2细胞的毒性。结果表明,以丝裂霉素为阳性对照,所用剂量范围内SVC Ⅱ(4-20μg/ml)能抑制HEp-2细胞生长和有丝分裂,对人癌细胞的毒性与药物剂量和作用时间成正相关,其中大剂量的SVC Ⅱ的抑制作用略强于MMC,提示SVC Ⅱ是蝎毒中具有抗癌活性的细胞毒素,可望成为来源于中药的新抗癌药。
Cytotoxicity assay, tumor cell killing assay, colony formation assay and growth inhibition assay were used to detect the toxicity of Buthus martensil karsch scorpion venom fraction Ⅱ (SVC Ⅱ) to human sore throat HEp-2 cells. The results showed that with mitomycin as positive control, SVC Ⅱ (4-20μg / ml) could inhibit the growth and mitosis of HEp-2 cells in a dose range. The toxicity to human cancer cells was positively correlated with the dosage and duration of action, The inhibitory effect of high dose SVC Ⅱ was slightly stronger than that of MMC, suggesting that SVC Ⅱ is a cytotoxic agent with anticancer activity in scorpion venom and could be expected to become a new anticancer drug derived from traditional Chinese medicine.
